Abstract

Danau Air Asin Gili Meno merupakan satu-satunya danau hipersalin di Pulau Lombok, Provinsi Nusa Tenggara Barat, yang memiliki potensi sebagai habitat berbagai mikroorganisme ekstremofilik, termasuk bakteri halofilik. Bakteri halofilik merupakan mikroorganisme yang mampu tumbuh pada lingkungan dengan konsentrasi garam tinggi dan berpotensi menghasilkan berbagai senyawa bioaktif serta enzim yang bernilai ekonomi, seperti amilase, protease, dan nuklease, sehingga berpeluang dimanfaatkan dalam bidang bioteknologi, industri, dan lingkungan. Namun, informasi mengenai keanekaragaman bakteri halofilik di Danau Air Asin Gili Meno masih sangat terbatas. Penelitian ini bertujuan mengisolasi dan mengidentifikasi bakteri halofilik yang berasal dari Danau Air Asin Gili Meno, Kabupaten Lombok Utara. Penelitian menggunakan metode isolasi bakteri pada media Nutrient Agar yang diperkaya dengan air asin steril, kemudian dilakukan karakterisasi berdasarkan morfologi koloni, pewarnaan Gram, serta uji fisiologis dan biokimia. Hasil penelitian menunjukkan bahwa isolat Spl A1, Spl B1, Spl C1, Spl D1, Spl E1, dan Spl E2 yang berasal dari Danau Air Asin Gili Meno, serta isolat Spl F1 dari Pantai Gili Meno, memiliki karakteristik yang mengarah pada genus Halobacterium sp., yaitu kelompok Archaea halofilik yang mampu tumbuh pada lingkungan dengan salinitas tinggi. Hasil penelitian ini memberikan informasi awal mengenai keberadaan bakteri halofilik di Danau Air Asin Gili Meno sebagai dasar pengembangan penelitian biodiversitas mikroorganisme ekstremofilik dan pemanfaatannya dalam bidang bioteknologi.
